The OVERLORD-MS trial, conducted across Norway and Sweden, compared two B-cell depleting therapies, rituximab and ocrelizumab, in patients with newly diagnosed relapsing MS. Over 30 months of follow-up, the study delivers important insights with both clinical and global health implications.

Key findings

The trial showed that:

  • Rituximab was as effective as ocrelizumab in controlling disease activity over 30 months 
  • Safety profiles were comparable between the two treatments

Part of the EBV-MS research initiative

Importantly, the OVERLORD-MS trial is embedded within the EU-funded EBV-MS project, which investigates the role of Epstein–Barr virus (EBV) in the development and progression of multiple sclerosis.

Because both rituximab and ocrelizumab target B cells, the main reservoir for latent EBV infection, the study also provides valuable biological material. These samples are now being used to explore how B-cell depletion influences EBV-related immune mechanisms in MS, helping to bridge clinical outcomes with disease biology.
